Application of antibody-liposome-Rh2 in periodontitis via maintaining bacterial homeostasis

As a keystone pathogen in periodontitis, Porphyromonas gingivalis (P. gingivalis) can not only induce the blooming of inflammophilic microbes but also become dormant to resist antibiotics. Periodontitis is more intractable than infectious disorders caused by typical species, and antibiotics are not effective. Targeting-factor interventions are needed to destroy as keystone pathogen and collapse its inflammophilic flora. Therefore, we developed a targeting nanoagent: antibody-conjugated liposomal drug carriers with ginsenoside Rh2 (A-L-R) for pleiotropic benefits. A-L-R showed high quality in HPLC, FTIR, TEM detection. With live/dead cell staining and in a serious of antimicrobial effects assay, A-L-R could affect P. gingivalis only. With FISH staining and in PMA-qPCR assay, the clearance of P. gingivalis by A-L-R was more than other groups and only the proportion of P. gingivalis was reduced by A-L-R in monospecies culture. Moreover, A-L-R could target P. gingivalis with high efficiency and low toxicity in vivo. A-L-R could maintain homeostasis with a relatively stable oral microflora, to achieve a more precise sterilization effect. Accordingly, this targeting nanomedicine can offer the new strategies for periodontitis therapy, providing a foundation for the development of periodontitis prevention and treatment.
