The present invention relates to an isolated cell or cell line comprising an endogenous genome and an exogenous nucleic acid molecule integrated into the endogenous genome， the exogenous nucleic acid molecule comprising a multiple insertion site， said multiple insertion site comprising three or more different paired target sites for pairs of zinc finger nucleases， wherein the paired target sites are situated such that pairs of the zinc finger nucleases bind and cleave as heterodimers， and wherein the paired target sites are not present in the endogenous genome of the cell or cell line， the cell or cell line further comprising a donor sequence integrated into the multiple insertion site following cleavage by a pair of zinc finger nucleases that binds to the paired target site. Disclosed herein are methods and compositions for targeted integration and/or targeted excision of one or more sequences into a cell， for example， for expression of one or more polypeptides of interest.
