The iPSC-derived pulmonary epithilial cells in micro-patterned culture plate [scRNA-seq]

We established a system of generating alveolar or airway organoids in a micro-patterned culture plate. In this system, various lung cell types were identified, depending on the culture condition. Overall design: Human iPSC-derived lung progenitor cells were seeded onto micro-patterned plates and cultured in "DCIK+3i" (CHIR99021, SB431542 and Y27632) medium for 7 days to induce alveolar organoids involving alveolar type 2 cells in the DCIK+3i medium. Then, the culture medium was switched to DCI+LATS(LATS-IN-1) medium to induce AT1 cells or "PAL" medium to induce alveolar transitional state cells (iATCs), respectively. The organoids in each condition were dissociated using Accutase and subjected to single-cell RNA sequencing analysis.
