We previously have found that RIPK3 induces robust anti-viral and inflammatory transcriptional signaling in primary cortical neurons, however the complete molecular mechanism governing this pathway is incompletely understood. Here we show RIPK1 is required to induce RIPK3-dependent transcription in ZIKV-infected neurons. Primary cortical neurons that were heterozygous (WT-RHIM) or homozygous (RHIM-RHIM) for a mutation in the RHIM domain of RIPK1 were infected with Zika virus (ZIKV) for 24hrs (n=4/group). Neuronal RNA lysates were then submitted for bulk RNAsequencing.
