Demonstrate the feasibility of using flow cytometry to sort cells based on epigenetic modification levels, namely histone 3 lysine 9 trimethylated (H3K9me3) and DNA methylation (5mC)) by using bimolecular fluorescence complementation (BiFC) probes couple to FACS. Conclusion: we developed a platform technology to analyze and sort single cells based on their epigenetic modification levels, specifically H3K9me3 and 5mC. Cells can be analyzed using FACS to reveal their intrinsic heterogeneity in epigenetic modifications. The sorting capability developed by our novel sensors allows us to unequivocally dissect epigenetic contributions to tumor spheroid growth and the development of drug resistance.
