RNA-FISH of Xist in iXist-ChrX mouse embryonic stem cells upon acute degradation of YY1

Here we provide the raw RNA-FISH images displayed in the study "YY1 binding is a gene-intrinsic barrier to Xist-mediated gene silencing". These images were collected from iXist-ChrX mouse embryonic stem cell lines engineered with the FKBP12 degron tag fused to endogenous YY1, such that addition of dTAG-13 acutely degrades YY1 protein from cells. Degradation of YY1 - either prior to Xist induction or after prolonged Xist induction - has little effect on the appearance of Xist RNA territories over the Xi chromosome.
