We engineered KEAP1 VUS and annotated oncogenic variants using prime editing. We used a transcriptional antioxidant response element (ARE) reporter to test the ability of these variants to induce KEAP1 LOF and thus activate NRF2-dependent transcription. Moreover, we used the prime editing sensor approach, wherein a synthetic version of the endogenous prime editing guide RNA (pegRNA) target site (the "sensor") is included adjacent to the pegRNA. The sequencing contained in this repository contains the amplicon sequencing of the pegRNA-sensor cassettes from DNA extracted from FACS-sorted samples, which were sorted into 4 bins on the basis of their GFP expression (the fluorophore linked to the ARE reporter).
