A/C plasmids are an important group of broad-host-range plasmids that drive the spread of antibiotic resistance genes in enteric human pathogens. In particular, these plasmids are linked to the emergence of Carbapenem resistant Enterobacteriaceae (CRE) due to their carriage of genes encoding resistance to extended spectrum cephalosporins and last-line carbapenems. In this project, hyper-saturated transposon mutagenesis coupled with transposon directed insertion site sequencing (TraDIS) were utilised to determine the complement of genes required for conjugation of A/C plasmids. The plasmid mutant libraries were conjugated from E. coli TOP10 cells to E. coli J53 cells. The data in this project are TraDIS reads from two independently constructed pMS6198A-Tn5 libraries before and after conjugation.
