Materials and Methods A fresh heart extracted from a female bovine (age: 1 year and 6-7 months, weight: 140-150 kg, time of death: 11:00, heart removal time: 11:40) was delivered on ice to the Orbeli Institute, and dissection started at 12:40 AM (Photo_Heart_Disection.jpg). Pieces of tissue were extracted from the left atrium. Four left atria tissue pieces were used for executing several RFA lesions and imaging. RFA machine (EPT-1000 XP, Boston Scientific) was set up at 8 W power and 15 s duration. Although, in some thin parts of the tissue the ablation is interrupted before, as follows. White light image was taken under illumination with white LED light. While imaging, the samples were immersed in cold PBS together with ice (as in Photo_HSI_Acquisition.jpg). Also, sample 4 was imaged both on Dec 5, 2024 and Dec 6, 2024, for evaluating sample conservation. Hyperspectral images were taken with a Nuance FX Imaging System (model: N-MSI-FX, CRi, Woburn, MA, USA). For each sample, several cubes were acquired under illumination with each of the wavelengths above mentioned, as described in the respective metadata files for each sample: Metadata_Sample1.csv Metadata_Sample2_Part1.csv Metadata_Sample2_Part2.csv Metadata_Sample3.csv Metadata_Sample4_Day1.csv Metadata_Sample4_Day2.csv Note: During the acquisition of Sample2 a problem with instrument computer ocurred, leading to a change in the binning/ROI settings for this 4D cube. Briefly, the Sample2_Part1 has a 348x256 pixels size, while the Sample2_Part2 has a 696x520 pixels size. Besides, as the camera distance/focus to the sample was the same, this images can be matched by applying a binning factor of 2 and cropping of 4 pixels at the bottom to Sample2_Part2 files. Also, the scale picture was taken using the same setup of Sample2_Part1 and the obtention of the pixel size for Sample2_Part2, Sample3 and Sample4_Day1 files can be made applying an approach similar (but inverse) to the previously mentioned. TTC Staining After HSI acquisition, the ablation on the samples were revealed with 2,3,5-Triphenyltetrazolium chloride (TTC) staining. TTC solution was prepared the same day at 1% concentration. Two slices were cut from the center of each lesion, and immersed in TTC solution. After at least 20 minutes of incubation, slices were photographed together with scale, for estimating lesion size.
