We report here with this study the genome-wide DNA methylation level analysis at single-nucleotide resolution by Next Gen bisulfite-sequencing using the Methyl-MaxiSeq platform from Zymo Research Services (Irvine, CA) in plants overexpressing the gene 5-Methyltetrahydropteroyltriglutamate Homocysteine Methyltransferase1 (METS1) comparatively to Col-0 plants both basal conditions and following Pseudomonas syringae DC3000 infection. Overall design: We analyzed whole-genome DNA methylation level in the three methylation contexts CG, CHG and CHH (Methyl-seq) in Col-0 and 35::METS1#1 plants, and both under basal and P.s. DC3000-inductive conditions, using three biological replicates for each condition. The results from this analysis were confirmed for the most relevant condition (bacteria inoculation) with 35::METS1#2 plants.
