Primary human preadipocyte differentiation using bulk RNA-sequencing

We performed bulk RNA-sequencing on primary human preadipocytes in which we induced differentiation, and collected samples at 6 timepoints across 14 days. The purpose of our study was to study gene expression during the course of preadipocyte differentiation into adipocytes. Overall design: We induced a 14 day differentiation in cryopreserved human subcutaneous primary white preadipocytes. We collected RNA at the 0, 1, 2, 7, and 14 day timepoints with four replicates at each time point. RNA-sequencing libraries were prepared using the Illumina TruSeq Stranded mRNA kit and sequenced on one lane of an Illumina NovaSeq S1 flowcell to produce an average of 42 million reads per sample.
