Extracellular matrix (ECM) acts as an intrinsic and natural microenvironment of cells, has been shown a pivotal effect on numerous biological processes of MSCs. Especially, MSCs are inclined to differentiate into specific cells lineage commitment when grow on their adaptive and tissue-like microenvironment elasticity. The mechanism still remains elusive on mechanical transduction processes in the microenvironment. In this experiment, human bone marrow mesenchymal stem cells were seeded on substates with different ECM rigidity, and the changes of gene expression were detected by chips, so as to explore the effect of extracellular microenvironment on stem cell differentiation. Human bone marrow mesenchymal stem cells were seeded on substrates of 32Kp and 0.5Kpa, and the total RNA samples were collected on 3 and 7 days respectively for chip detection.Therefore, the test samples were divided into 4 groups according to different culture time and matrix hardness (3H-represents the experimental group cultured on 32kpa substrate for 3 days, 7H-cultured on 32kpa substrate for 7 days, 3S-cultured on 0.5kpa substrate for 3 days, and 7S-cultured on 32kpa substrate for 7 days)
