This project aims to determine the in-vitro antimicrobial susceptibility patterns of multidrug-resistant (MDR) Gram-negative ESKAPE pathogens using the VITEK 2 Compact system. The primary objective is to molecularly characterize antibiotic resistance genes, virulence factors, and mobile genetic elements in MDR bacterial variants obtained from collected samples. This will be done through whole genome sequencing (WGS) data analyzed using bioinformatics tools.We will study capsular types, plasmids, insertion sequences, and phage regions associated with these pathogens. Additionally, we will identify plasmids linked to antimicrobial resistance (AMR) determinants and investigate Clustered Regularly Interspaced Palindromic Repeats (CRISPR) regions for genotyping MDR bacteria.This research seeks to enhance our understanding of the molecular biology of MDR bacteria and identify novel antibiotic target genes or proteins. Furthermore, we will analyze WGS data variants to highlight the genetic differences and international spread of antibiotic-resistant organisms, particularly comparing global databases with data from Turkey.Overall, this study aims to provide insights into the resilience and adaptability of MDR ESKAPE pathogens.
