Expression of Physcomitrium patens PPR78 in Escherichia coli

The moss C-to-U RNA editing factor PPR78 of Physcomitrium patens is expressed in N-terminal fusion with His6-MBP in Escherichia coli Rosetta2DE3 cells. A specific editing target is co-transcribed downstream of the protein coding sequence. Expression is induced by the addition of IPTG. Total RNA is isolated after 20h of incubation in selective LB media containing 0.4mM zinc to screen for off-targeting of PPR78 in the bacterial transcriptome.
