Resveratrol inhibits migration, invasion and EMT of bladder cancer 5637 cells through suppression of TGF-β signaling pathway

AimTo study the effects of resveratrol (RSV) on migration and invasion of bladder cancer 5637 cells as well as the possible mechanisms.MethodsSulforhodamine B (SRB) assay was used to detect the proliferation of 5637 cells. Transwell method was used to examine the migration and invasion of 5637 cells. Gelatin zymography was employed to detect the matrix metalloproteinase-2/-9(MMP-2/-9) secretion by 5637 cells. Western blot was applied to determine the expressions of marker proteins of EMT and associated signal molecules such as Smad2/3, p-38 MAPK, Akt, ERK.ResultsTreatment with RSV for 24, 48 and 72 h inhibited the proliferation of 5637 cells significantly. RSV at the concentrations of 13. 75, 27. 5, 55, 110 μmol·L-1 inhibited the migration, invasion and MMP-2 secretion of 5637 cells. Western blot analysis indicated that RSV could reverse the transforming growth factor-β (TGF-β)-induced EMT and the phosphorylation of Smad2/3, p-38 MAPK and Akt.ConclusionRSV inhibits the migration, invasion, secretion of MMP-2 and TGF-β-induced EMT of bladder cancer 5637 cells through suppression of TGF-β signaling pathway.
