We analyzed effects of IFNAR deficiency on immune cells of the GBM TME and also analyzed how anti-PD1 affected these immune cells in absence of type I IFN signaling. We also analyzed the effects of anti-PD1 treatment on the immune cell populations in absence of IFN production. CD45.2+ tumor-infiltrating immune cells were sorted from brains of WT and IFNaR-/- mice 20 days following Gl261 intracranial injection. In addition, in order to analyze the effects of ICB therapy on immune cells in absence of IFN signaling, anti-PD1 treatment was intraperitoneally injected at a dose of 200 ug/mouse on days 7,11,15,19 following Gl261 injection. CD45.2+ immune cells were sorted from the brains of anti-PD1 treated WT and IFNaR-/- mice 25 days following Gl261 injection.
