Purpose: To assess changes in gene expression profiles of mouse ovarian gruanlosa cells from normal control mice, 11-month mice and 11-month mice treated with CEFFE. Methods:The control group comprised 8-week-old female mice, and the 10-month-old female mice were equally divided into two groups, Aged and Aged +CF groups. The mice from Aged +CF group were treated with 200 µL CEFFE (protein concentration: 3 µg/µL) via tail vein every two days for 2 weeks. At the same time, mice from Control and Aged group were injected with the same volume of saline. Then at the forth week ,ovaries were collected. Furthermore, the mRNA-Seq of mouse ovaries were performed to explore underlying mechanism of CEFFE. Results: 1302 genes were significantly increased and 1425 genes were significantly decreased in expression level between Aged vs Ctrl gorup.154 genes were significantly increased and 167 genes were significantly decreased in expression level between Aged+CF vs Aged gorup. Overall design: total RNA was isolated and sequenced from the mouse ovaries from normal control mice, Aged mice and Aged mice treated with CEFFE in triplicate using an Illumina high-seq 2500. Raw data was analyzed using TopHat. Genes were considered changed which demonstrated fold-change>=1.5, and p value <=0.05.
