Whole transcriptome of the wild-type PRV isolate (strain PRV-MdBio) was analyzed in different time points of infections. The virus was propagated on three different cell lines: porcine kidney epithelial cell line (PK-15), C6 cell line derived from a rat glial tumor, PC-12 cells derived from a pheochromocytoma of the rat adrenal medulla and NRK cells derived from rat kidney cells. Poly-A selected viral RNA was isolated in the following time points post-infection: 1h,2h,4h,6h,8h,12h in triplicates. Direct cDNA from each time points and dRNA from a mixture of time points was sequenced on Oxford Nanopore MinION sequencer device. The comparative transcriptome analysis enables temporal differentiation of viral and host cell gene expression.
