We developed a sensitive splinted ligation-based and 5-phosphate independent library preparation protocol (SLiPiR-seq) to analyze plasma cell-free RNA. We show it outperforms conventional adapter ligation-based method in detecting multifarious types of RNA in blood. Raw sequencing reads used for technology optimizations of SLiPiR-seq are provided.
