High throughput PIXUL-ChIP-seq genome-wide study allows to map simultaneously Pol II peaks in multiple organs harvested from the same mouse. Hearts, kidneys, livers and lungs harvested from two mice were simultaneously sheared in a 96-well plate using PIXUL, high throughput chromatin sonication instrument. PIXUL-sheared heart, kidney, liver and lung chromatin was then used in multiplex microplate ChIP platform (Matrix ChIP) to analyze in parallel genome-wide binding of Pol II in these mouse organs. Indexed libraries were constructed from ChIP DNA using Tn5 transposase tagmentation. The eight libraries were pooled and clustered onto a flow cell lane. Sequencing was performed using an Illumina HiSeq 2500 in Rapid Mode employing a paired-end read, 50-base read length (PE50) sequencing strategy.
