For this study, Chinese Hamster Ovary (CHO) cells (221-107; mAb4/anti-MUC1/IgG1? cell line) were grown in a 2 L fed-batch and sampled for 15 days (D0 to D14). The hypothesis tested was: 'lower glutamine levels result in lower lactate levels in fed-batch cultures without affecting mAb productivity'. These cells were fed 2% Feed 7a and 0.2% Feed 7b once daily from D2-13. For high glutamine conditions, 0.5% of 200 mM glutamine was fed daily between D2-13. For low glutamine conditions, glutamine was only fed if levels dropped below 0.22 g/L. For either condition, if glucose levels dropped below 4 g/L, glucose was fed to 5 g/L with a 500 g/L solution. There were eight high glutamine (8 mM) bioreactors and four low glutamine (2 mM) bioreactors. Genomics, transcriptomics, metabolomics and proteomics were generated for this study. For the transcriptomics, samples were taken at eight time points (D4, 5, 6, 7, 8, 11, 12 and 14) and short read sequencing was performed using Illumina NovaSeq 6000 system. Transcriptomics was analysed by Edinburgh Genomics (UoE).
