To screen the transcripts from Th. elongatum 7EL, we inoculated the spikes of translocation line Zhongke 1878 with Fusarium graminearum. We performed full-length transcriptome sequencing after 96 hours. To detect the expression level, we performed RNA-Seq on Zhongke 1878 after 12h, 24h, 48h and 96h. Samples from different spikes of Zhongke 1878 were used in the study. The high-quality, full-length cDNAs was synthesized from total RNA by using a SMARTer™ PCR cDNA Synthesis Kit by Berry Genomics Co. Ltd, China. Sequenced transcripts were mapped to Chinese Spring reference genome using the software GMAP. The unmapped transcripts were extracted. For next generation sequencing, the cDNA libraries were constructed following Illumina standard protocols and sequenced with Illumina HiSeq 2500 by Berry Genomics Co. Ltd, China. Sequenced reads were trimmed using Trimmomatic (version: 0.36), then mapped to the screened transcripts using hisat2 (version: 2.0.0-beta) and the FPKM value were calculated using StringTie (version:1.3.3).
