Effect of Qingyi Decoction on the Treatment of Acute Pancreatitis Mice via mRNA Sequencing Analysis

Acute pancreatitis (AP) is an inflammatory disease of pancreatic tissue caused by pancreatic acinar cell injury. AP treatment of Qingyi Decoction(QYD) is a widely and commonly used traditional Chinese medicine prescription. A mice model of acute pancreatitis was established and treated with Qingyi Decoction, and mRNA high-throughput sequencing was performed on the pancreatic tissue of mice treated with Qingyi Decoction for the first time. There were 4477 and 4359 differentially expressed genes in the control group compared with the model group and the Qingyi Decoction group, while there were 8 differentially expressed genes in the model group compared with the Qingyi Decoction group by analyzing the mRNA expression profiles. Analysising of KEGG, GO, GSEA and network interaction, it was found that its therapeutic effect was closely related to apoptosis, inflammatory response and autophagy process. Overall design: Wild-type C57BL/6 mice were randomly divided into three groups. Each mouse was given once per hour for 7 intraperitoneal injections.Model group and Qingyi Decoction group were injected 50 mg/kg caerulein,and LPS was added for the last time. Control group was injected with the same amount of normal saline. Qingyi Decoction group was given medication at 0 and 8 hours after modeling,while control group and model group were treated with saline. Pancreatic tissue was taken for high throughput sequencing of mRNA 24 hours after the start of modeling.
