RITA, Nutlin3, and CTX1, inhibitors of p53-MDM2 interaction, could increase the CRISPR/Cas9-mediated precise genome editing in sheep fetal fibroblasts (SFFs). To detect the potential genotoxic activity of these three small molecules, we performed whole genome sequencing (WGS) in the Cas9/sgRNA/HDR/MDM2 antagonists treated SFFs, and observed that the number of single-nucleotide polymorphisms (SNPs) and indels did not show obvious increase compared to the DMSO-treated control group following treatments with RITA, Nutlin3, and CTX1 alone or small molecules mixture.
