In this study, we cloned a gene encoding GSNOR and employed genetic approaches to investigate its role in this aflatoxigenic fungus. Our findings revealed that GSNOR deletion increased NO and ROS levels, making the strain more sensitive to NO stress and oxidative stress. We also found that GSNOR deletion impaired asexual development, notably attenuated virulence on kernels and decreased aflatoxin production. Our study highlights the essential role of GSNOR in fungal development, secondary metabolism, and virulence of <i>A. flavus</i>.
