ATF6ais required for the inflammation and survival in rheumatoid arthritis fibroblast-like synoviocytes

To characterize transcriptome changes upon ATF6a knockdown by the siRNA in rheumatoid arthritis fibroblast-like synoviocytes (RA FLSs) Overall design: RA FLSs were isolated from the synovial tissue of RA patient who underwent knee joint replacement surgery. FLSs of passages 4 were used in this study. The RA FLSs seeded in six T75 culture flasks were classified into two groups (each group with triplicates): group 1 with negative control siRNA transfection (the siNC group), group 2 with ATF6a siRNA transfection (siATF6a group). RA FLSs transfected with either siNC or si ATF6a were trypsinized and collected 72 hours post transfection. Total RNA was extracted to conduct cDNA library construction for RNA transcriptome sequencing.
