Complex Rearrangements in RPE1

Here we report a methodology for the efficient generation of complex SRs in vitro, based on perturbing karyotypically normal cells and employing a selection barrier, before analyzing transformants by massively parallel sequencing. We employed this methodology to characterize mechanisms of catastrophic SR events, the temporal procession of their formation as well as the resulting consequences on the genome.
