MNase sequencing of macronuclei from vegetative Paramecium tetraurelia

Aims: Characterizing the distribution of nucleosomes in Paramecium tetraurelia. Materials & methods: Mono-nucleosomal DNA fragments were obtained by MNase digestion of chromatin isolated from macronuclei of exponentially growing vegetative P. tetraurelia (strain 51 mating type 7) cells for 2 biological replicates (PTET_WT_VEG_MNASE_SEQ_MonoNuc_3-2 and PTET_WT_VEG_MNASE_SEQ_MonoNuc_4-1). As controls, two DNA samples extracted from macronuclei of exponentially growing vegetative P. tetraurelia (strain 51 mating type 7) were digested with MNase and DNA fragments of the same molecular weight as the mono-nucleosomal DNA fragments were selected for library preparation (PTET_WT_VEG_MNASE_SEQ_NakedDNA_D1 and PTET_WT_VEG_MNASE_SEQ_NakedDNA_D2). Paired-end Illumina sequencing was performed to assess the genome-wide nucleosome profiles. Results: Our results revealed enrichment of well-positioned nucleosomes downstream Transcription Start Sites.
