Microscopy-based assessment of H3K27-acetylation throughout sperm precursor formation in fruit fly testes

This repository contains the data and MatLab analysis scripts of the analysis of histone 3 lysine 27 acetylation (H3K27ac) and recruited RNA polymerase II (Pol II Ser5P) levels throughout sperm precursor formation in fruit fly testes. Samples were prepared by Agnieszka Pancholi in the laboratory of Sylvia Erhardt. Images were recorded by Marcel Piepers in the laboratory of Lennart Hilbert. Image analysis scripts were developed and images were analyzed by Lennart Hilbert. To analyse date data, the raw image data are first extracted into MatLab-native files using the MultiPosition_extraction_czi_lazy.m script. The actual analysis is then carried out using first the ClusterAnalysis_revisions_Mar2026.m script. The extracted data can be reviewed using the ReviewExtractedStacks.m script.Several images needed to be excluded from analysis because: the apical hub could not be clearly distinguished, the sample was too warped to reliable establish a spatial coordinate representing precursor formation, the recorded data was corrupoted. A list of excluded files is deposited for reference in the file QA_SampleRemoval.pdf.
