We investigate the effect of altered kinase levels (ZAP70) in the VDJ usage of B-cell receptors in humanized mice Human cord blood derived CD34+ progenitor cells were transduced with a GFP vector control, or GFP-labelled ZAP70 lentivirus and injected into NOD/Scid/Il-2rg/Kit-W41J/Tyr mice. GFP positive and negative naïve B-cells (CD19+CD27-CD10-) were isolated from 6-month old mice by FACS. 10,000 cells pooled from 3 independent mice per group were subjected to single cell BCR VDJ sequencing (10X).
