This study aims to develop a synergistic nanotherapy (Curcumin-FGF21 dual-loaded nanocomplexes) to simultaneously target two key aspects of NASH pathology: liver signaling pathways and gut microbiota. We intend to demonstrate that this therapy can improve hepatic metabolic disorders, oxidative stress, and inflammation by coordinately regulating the FXR/Nrf2/NF-kappa B signaling axis, while also modulating the intestinal microbiota to reduce its detrimental impact. The goal is to achieve effective hepatoprotection in a diet-induced mouse model of metabolic dysfunction-associated steatohepatitis (MASH/NASH). This work seeks to provide a novel multi-target, multi-mechanism integrated therapeutic strategy to overcome the current limitations in NASH treatment.
