This study has revealed the effect of melatonin on gene expression and mineralization of ameloblastic lineage cells (ALCs) as well as a potential mechanism through melatonin receptor-mediated JNK3-Arrb1 signaling pathway. The main finding of this study indicates that melatonin promotes an in vitro ameloblastic mineralization of ALCs through melatonin receptors based on specific ameloblastic gene and protein expression, ALP activity and Alizarin Red S. RNA-seq analysis also showed the up-regulation of neural JNK3 and down-regulation of Arrb1 in ALCs during melatonin induction. This is also confirmed by the inhibition of phosphorylated JNK3 deficiency to delay the mineralization, and Arrb1 overexpression down-regulated nuclear JNK3.
