Single cell analysis of newly diagnosed AML patients

Acute Myeloid Leukemia (AML) is a heterogenous disease characterized by immature blasts at different maturation stage. We used single cell sequencing technique to analyze newly diagnosed AML patients. Bone marrow biopsies were routinely collected before treatment initiation. Samples were freshly frozen with freezing medium containing 20% fetal calf serum (FCS) and 10% dimethyl sulfoxide (DMSO) in Dulbecco’s modified Eagle medium (DMEM) and stored in liquid nitrogen. All frozen BM samples were retrieved immediately before sample processing. The 5 prime gene expression libraries were prepared using a 10x Genomics Chromium Controller instrument and Chromium Single-Cell 5 V5.1 reagent kits (10x Genomics).
