Inhibition of PC reverse metformin resistance pancreatic cancer

Using lentivirus infection PDAC cells to construct PC stably knocking down cell lines and detecting its effect on PDAC progression in vivo and vitro. RNA-sequencing analysis on PC knocked down in PDAC cells and the results were verified by RT-qPCR. Seahorse XF flux analyzer used to analyze mitochondrial oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) changed with or without metformin. The therapeutic efficiency of metformin combined with PC interference in PDAC were evaluate in vitro. Overall design: Comparative gene expression profiling analysis of RNA-seq data for PDAC cells (n=3) and its PC stably knocking down cell lines(n=3).
