BRAF V600E has been shown to be resistant to both Cetuximab and Dabrafenib used alone. Clinical trials to assess the combination and addition of the MEK inhibitor Trametinib have shown favourable results. We therefore seek to model and understand the molecular mechanisms of resistance to this combination therapy before the development of resistance in the clinic. This lentiviral mutagenesis mediated forward genetic screen will enable us to identify overexpression events on a genome wide scale which engender resistance to this therapy.
