H3.3 phosphorylation promotes high levels of histone acetylation in mouse embryonic stem cells, which are central to the initiation of new transcription during lineage specification. Overall design: Native ChIP analysis of H3K27ac in several mouse embryonic stem cell lines (Control and H3.3KO, HIRAKO, ATRXKO, DAXXKO). Native ChIP analysis of H3K64ac, H3K122ac, H3K4me1 in two embryonic stem cell lines (Control and H3.3KO). Crosslinking ChIP analysis of H3.3 (Control, ATRXKO and DAXXKO) and p300 (Control and H3.3KO). Native ChIP analysis of H3K27ac in embryoid bodies at Day 4 of differentiation (WT and H3.3KO and addback cells). All inputs sequenced.
