Single-cell sequencing of diaphragms from control, SRSF1 knock-out, and SRSF2 knock-out mice revealed distinct cell types and highlighted the different roles of SRSF1 and SRSF2 in the development and function of skeletal muscle satellite cells. Overall design: The diaphragms were isolated from one-day-old control, SRSF1 KO, and SRSF2 KO mice. The knockout of SRSF1 or SRSF2 was driven by Cre recombinase under the control of the MyoD1 promoter. To obtain a single-cell suspension, the diaphragms were fully digested with Dispase I and Collagenase type XI. The resulting single cells were then used for single-cell sequencing.
