Here we aim to dissect the complex nature of the acute systemic response of HSCs to lipopolysaccharide (LPS), mimicking a bacterial infection. Acute LPS challenge transiently induced proliferation of quiescent HSCs in vivo, without yet impairing HSC function. We hypothesized that the effect of LPS on HSCs is indirectly mediated via proinflammatory cytokines. Our results showed that Acute LPS exposure depends in part on IFN signaling to activate HSCs. Overall design: Bone marrow (BM) was isolated from mice 18h post treatment of PBS (control), LPS or IFNa treatment. This experiment included 6 biological replicated per condition. BM cells were then subjected to lineage depletion followed by sorting of Lin- ckit+ CD150+ CD48- HSCs. The RNA was then isolated from the sorted cells and hybridized on Illumina chips.
