miRNA profiling in mouse peritoneal macrophages from macrophage-specific NFATc3 transgenic mice and their control littermates

We generated NFATc3 macrophage-specific transgenic mice (NFATc3MTG) using the piggyback Transposon. Peritoneal macrophages were isolated from NFATc3MTG mice and their control littermates (wild-type), and the miRNA expression profile was analyzed with deep sequencing. NFATc3MTG and wild-type mice (10-weeks old) were injected with 1 mL 3% thioglycolate. Peritoneal macrophages were isolated via peritoneal lavage 4 days later and cultured in medium for 24 h. Cells were harvested, RNA isolated, and miRNA sequencing performed using the Illumina HiSeq. miRNA library was built using NEBNext® Multiplex Small RNA Library Prep Set for Illumina® (New England Biolabs, Beverly, MA) and run on the HiSeq 2500 (Illumina) in 50 bp single-end reads.
