The goas of this experiment were: 1) to assess the effect of CART19 killing on tumors dissected from xenograft models of B-NHL; 3) to assess the effect of combining tazemetostat/CART19 on tumors dissected from xenograft models of B-NHL. OCI-Ly18-GFP+ cells were sub-cutaneously implanted into NSG mice. 3 days later, mice were treated with tazemetostat (150 mg/kg/daily) or vehicle for 14 days. Mice were then infused with CART19 cells or UTD. 11 days post CART19 infusion, mice were sacrified, tumor were dissected and processed to prepare a single cell suspension. Cell sorting separated tumor-GFP+ cells from CART19 cells. Total RNA was extracted from tumor cells and submitted for bulk RNA sequencing.
