HoxA9-Meis1 transformed mouse myeloid cell lines were treated with the antiviral agent Beta-d-N4-hydroxycytidine (NHC/EIDD-1931). Single cell cloning was performed and genome sequencing used to survey patterns of somatic mutations. Vehicle treated control samples were used for comparison. The parental cell line was assessed and compared to derivative lines that express exogenous uridine-cytidine kinase 2 (Uck2) or in which Uck2 has been inactivated by CRISPR-mediated editing. Three single cell clones were derived for each condition.
