1. CRISPR/Cas9 is often thought to allow precise genome editing, but the efficient and accuracy of sequence modification throughout genomic locations is barely understood. Here we present a large-scale dataset of 2,260 whole genome sequences of yeast strains receiving modifications from a pool editing experiment based a complex library of natural variants. 2. Four genomic locations were chosen for validating Cas9-induced structural variations (SV). Editing experiments were performed and single colonies were isolated and sequenced. 3. Six different editing systems were tested on their performance to minimize SV formation, on 9 distint loci. After editing, bulk gDNA samples were collected and sequenced.
