HIGH SEQUENCING DEPTH Head-to-Head CRISPRko CRISPRi Perturb-seq: Aligned, filtered data with guide assignments

Low MOI perturb-seq data (as Seurat objects) from parallel Cas9 CRISPRi benchmark library in K562 cells sequenced at Day 14 post-transduction. Fastqs were aligned with Illumina DRAGEN software, which filters droplets to "passing cells". Cells are further filtered to those with <15% mitochondrial transcripts and a single guide assigned. Guide assignments are stored in the metadata of each Seurat object. This data represents the second round of sequencing of these samples to yield improved sequencing depth. Each sample had their own two lanes on a Novaseq X 25B flowcell. The CRISPRko data has 31,572 mean transcriptomic reads per passing cell and the CRISPRi data has 28,559. While the entire experiment featured additional experimental time points and cell lines, only the Day 14 K562 samples were sequenced at this depth. The sample labeled "AC23" corresponds to the CRISPRko experiment. The sample labeled "AC24" corresponds to the CRISPRi experiment.
