In this study, we performed deep-mutational scanning of hotspot 1 and hotspot 2 of Fks1 and Fks2, both encoding beta-1,3-glucan synthases. Mutations in these hotspots often correlate with echinocandin treatment failure, therefore comprehensive mutational data can help predict echinocandin resistance. In combination we molecular dynamics simulations, we also use the data to unveil the molecular bases of cross-resistance and echinocandin-specific resistance, which can ultimately be used to re-design existing drugs.
