Peritoneal macrophages were isolated from WT or Hmgxb4 KO mice (Female mice, two mice each group). Macrophages were treated with or without LPS (100 ng/ml) for 6 hours, and total RNA was then extracted using TRIzol reagent (Invitrogen) and subjected to whole transcriptome RNA-seq analysis. Differential gene expression analysis was performed at the Genome Technology Access Center at Washington University. Ribo-Zero library was prepared for single-end sequencing in 50-bp length on a HiSeq 3000 system (IIIumina). RNA-seq reads were then mapped to Ensembl release 76 top-level assembly using STAR version 2.0.4b. The raw count for genes annotated in Ensembl GRCm38 were calculated with Subread:featureCount version 1.4.5.
