Mouse_Transposon_Validation

An in vivo tumorigenesis system that samples tissue and developmental contexts in the whole animal by using DNA transposons to deliver and stochastically express pools of candidate cancer gene cDNAs. Each transposon carries a promoter-less cDNA that traps promoters and enhancers as it translocates in the genome. If a transposon traps a promoter that expresses an oncogenic cDNA at an appropriate level, in a susceptible tissue, at a suitable developmental stage, so as to give a transformation phenotype with selective clonal advantage – tumorigenesis occurs. Exome sequencing uncovers cooperating mutations to the expressed cDNAs, and also insertional profiles of the tumour clones.
