The sampling site was located in the Meigu Dafengding National Nature Reserve, with a sampling depth of 10-20 cm. The sampling method involved selecting a 1x1 m subplot within a 20x20 m main plot and employing a five-point sampling technique to collect soil. At each selected point, after removing the litter layer, healthy roots were excavated with a shovel from the base of bamboo stems. Non-rhizosphere soil dislodged from the roots was collected. Gently shaking off the loose non-rhizosphere soil, the soil adhering to the root surface (approximately 2 mm thick) was taken as the rhizosphere soil sample. Rhizosphere soil collected from the five sampling points was combined into a single plastic bag and thoroughly mixed to obtain approximately 50-100 g of sample. A total of 72 samples were preserved at -80 degrees C for subsequent analysis of rhizosphere microorganisms.
