To identify TRPV1-associated molecular chaperones, we applied a PafA-based proximity labeling system using both N- and C-terminal TRPV1 fusions. Three groups were co-transfected with BCCP-Pup(E) for in-cell biotinylation. Biotinylated proteins were validated by immunofluorescence and Western blot, followed by streptavidin pull-down and mass spectrometry.
