To investigate the global gene expression change caused by suppression of csi-miR156a expression which reduced somatic embryogenesis in citrus, we compared gene expression profiles of MIM156 line (MIM156-L1) and the EV line (EV-L44) at 0, 15 and 30 days after somatic embryos induction (DAI). At 0 DAI, the callus of EV-L44 and MIM156-L1 have no different. At 15 DAI, the callus of EV-L44 changed from white loose cell clusters to yellow dense granules, and a few yellow structures resembling globular embryos appeared, whereas the morphology and color of MIM156-lines were not changed. At 30 DAI, the callus of EV-L44 was changed from light yellow to dark yellow, and a few green spherical embryos appeared, while the color of MIM156-lines remained white without the formation of spherical embryos. A total of 1880 differentially expressed genes were found between MIM156-L1 and EV-L44 at three stages. Our result indicated that suppression csi-miR156a expression could cause large-scale gene expression alteration.
