In this project, we performed GUIDE-seq in human 293T cells to identify CRISPR/cas9 gRNA-dependent off-target effects of gRNAs targeting the gamma-globin promoters through base editors. We coupled each gRNA with a Cas9 (SpCas9 or Cas9-SpRY) that corresponds to the Cas9 nickase version of the base editor used.
